The expression of TEAD1 in macrophages of myocardial tissue after acute myocardial infarction in mice
Objective To observe the expression and cellular localization of TEA transcription factor 1(TEAD1)in myocardial tissue of mice after acute myocardial infarction(AMI),and to explore the role and mechanism of TEAD1 in AMI.Methods Forty C57BL/6 mice(6-weeks-old)were randomly divided into AMI group and Sham operation group.After operation,the mice were fed with common diet for 1,3,7 and 14 days and sacrificed.In this study,HE staining and Masson staining were used to observe the patholog-ical changes of myocardial tissue after acute myocardial infarction.Immunohistochemistry was used to detect the expression of transcrip-tion factor TEAD1 and macrophage marker CD68 in myocardial tissue of mice after acute myocardial infarction.Immunofluorescence double-label method was used to observe whether TEAD1 was over-expressed in macrophages in myocardial tissue after acute myocar-dial infarction.Results Compared with the Sham group,the expression of TEAD1 and CD68 in AMI group was significantly increased(P<0.05),mainly in the infarction boundary area.In the AMI group,the expression of TEAD1 and CD68 showed a trend of first in-creasing and then decreasing over time,and reached the peak on the third day,and the differences between adjacent time points were sta-tistically significant(P<0.05).The results of immunofluorescence double labeling showed that the expression of TEAD1 in AMI group was higher than that in Sham group(P<0.05)except on the 14th day,especially on the 3rd day,and showed a trend of first increasing and then decreasing.The quantity difference of adjacent time points was statistically significant(P<0.05).Conclusion TEAD1 and macrophage marker CD68 are highly expressed in myocardial tissue after acute myocardial infarction,and TEAD1 can be localized in macrophages at the infarction boundary.
