Effects of LncRNA MEG3 overexpression on proliferation and EMT of Ishikawa cells in human endometrial carcinoma
Objective To investigate the effect of LncRNA MEG3 overexpression on the proliferation of Ishikawa cell in endo-metrial cancer and its role in the process of epithelial mesenchymal transition(EMT).Methods Ishikawa cells were divided into Control group(blank control),Len-nc group(transfection of lentiviral empty vector)and Len-MEG3 group(transfection of LncRNA MEG3 overexpression lentivirus).The expression levels of LncRNA MEG3,E-cadherin,N-cadherin,Vimentin and ZEB1 in each group of Ishikawa cells were detected by qRT-PCR,and scratch assay was used to detect the migration ability of cells in each group.CCK8 and clonal formation assay were used to detect the proliferation ability of cells in each group.Flow cytometry was used to detect the apoptosis of cells in each group.The Western blot method was performed to detect the expression levels of EMT-related proteins including E-cadherin,N-cad-herin,Vimentin and ZEB1.Result Compared with the Control group and the Len-nc group,LncRNA MEG3 expression levels were significantly increased in the Len-MEG3 group(P<0.05),the migration and proliferation ability of cells were decreased,and the apoptosis ability of cell was increased.While the expression of E-cadherin mRNA and protein was up-regulated and the expression of N-cadherin,Vimentin and ZEB1 mRNA and protein was down-regulated.Conclusion LncRNA MEG3 overexpres-sion inhibits the proliferation,migration ability and EMT process of Ishikawa cells in endometria cancer,while promoting apoptosis.
