miR-21-5p inhibited TLR4-induced cells proliferation of cutaneous squamous cell carcinoma
Objective To investigate the effects of TLR4 and miR-21-5p on the proliferation of cutaneous squamous cell car-cinoma and their targeting relationship.Methods TLR4 expression in skin squamous cell carcinoma tissues was detected by immuno-histochemistry,TLR4 in overexpressing cells and miR-21-5p in inhibiting cells were transfected by Toll-like receptor 4(TLR4)plasmid and miR-21-5p inhibitor,the relationship between TLR4 and miR-21-5p was analyzed by bioinformatics,and the relation-ship between TLR4 and miR-21-5p in skin squamous cell carcinoma was detected by RT-qPCR.Expression of TLR4 and miR-21-5p in cells,and the growth of A431 cells by CCK8,and the expression of TLR4 and the proliferation molecule ki-67 in cells by western bloting.Results TLR4 was expressed the highest in adjacent tissues of cutaneous squamous cell carcinoma,and its expression was lower in poorly differentiated tissues than in well differentiated tissues(P<0.05).TLR4 was expressed lower in A431 human cuta-neous squamous cell carcinoma cells than in HaCaT human immortalized keratinocytes(P<0.05).A431 cells inhibited the expression of miR-21-5p(P<0.05),and the inhibition of cell proliferation was detected,and the difference was statistically significant(P<0.05),TLR4 expression was reduced,the difference was statistically significant(P<0.05).Conclusion The mutual regulation of miR-21-5p and TLR4 affects the proliferation of A431 cells,and the interaction between miR-21-5p and TLR4 may play an important role in the cellular behavior of cutaneous squamous cell carcinoma.
