Study on the protective effect and mechanism of Ningxia lycium barbarum polysaccharide on photo-aging of UVB induced HaCaT cells
Objective To study the protective effect and mechanism of Ningxia lycium barbarum polysaccharide(LBP)on the photoaging induced by medium-wave ultraviolet(UVB)light in human immortalized keratin-forming cells(HaCaT).Methods The photoaging cell model was established by stimulating HaCaT cells with UVB-induced.Cell viability was determined by the multiple table tournament(MTT)method.The levels of interleukin-1β(IL-1β),interleukin-6(IL-6),and human tumor necrosis factor-α(TNF-α)affected by enzyme-linked immunosorbent assay(ELISA)were detected as an index to study the effects of different concen-trations of LBP on the anti-photo-aging activity of HaCaT cells and its mechanism.Results The proliferation activity of HaCaT cells decreased after UVB irradiation,and the cell activity of the irradiation group increased after intervention with lycium barbarum polysac-charide,and the cell survival rate was categorized into 95.49%and 100%when the LBP content was 80 μg/mL and 160 μg/mL..Com-pared to the blank control group and the UVB irradiation group,the LBP concentration was 160 g/mL,the levels of IL-1β,IL-6,and TNF-α were decreased significantly.Conclusion LBP has an anti-photoaging effects,the mechanism may be through inhibiting the proliferation activity of HaCaT cells by UVB induced and reducing the expression levels of IL-1β,IL-6 and TNF-α.
