[目的]为了发现具有抗烟草花叶病毒(TMV)活性的新化合物.[方法]通过取代、肼解、环化、取代4步反应将(噁)二唑片段引入到蓝刺头碱结构中,合成11个含1,3,4-噁二唑片段的蓝刺头碱衍生物,并对其进行了抗TMV活性测试.[结果]目标化合物结构经1H NMR、13C NMR和HRMS分析确证.抗TMV活性测试结果显示在质量浓度为500 mg/L时,8个蓝刺头碱(噁)二唑衍生物的抗TMV活性高于病毒唑,其中化合物5d和5j的活性最高.[结论]化合物5d和5j可作为抗TMV先导化合物进行进一步结构优化.
Design,synthesis and anti-TMV activities of echinopsine derivatives containing 1,3,4-oxadiazole fragment
[Aims]This study aims to discover novel compounds with high anti-TMV activity.[Methods]The fragment of 1,3,4-oxadiazole was introduced into echinopsine through 4 steps including substitution,hydrazine hydrolysis,cyclization and substitution to synthesize 11 echinopsine derivatives containing 1,3,4-oxadiazole,and their anti-TMV activity were tested.[Results]The structures of the target compounds were confirmed by 1H NMR,13C NMR and HRMS.The activity test results showed that the anti-TMV activities of 8 echinopsine 1,3,4-oxadiazole derivatives were higher than that of ribavirin at 500 mg/L,with compounds 5d and 5j having the highest activity.[Conclusions]Compounds 5d and 5j can be used as lead compounds for further structural optimization.
