无花果多糖分离纯化工艺及抗氧化性能研究

为更好地利用无花果副产物,提高其经济价值,探究了无花果多糖(FCW)抗氧化性能.采用微波辅助提取FCW,并对提取工艺进行优化.最佳条件为提取温度 70 ℃、料液比 1:40、微波功率 800 W和提取时间 50 min,在此条件下,FCW提取率 12.32%.以残渣模拟无花果生产后的废料,采用碱液浸提法提取无花果残渣多糖(FCA),并分析其热稳定性.结果表明,FCA在 260 ℃以下具有良好的热稳定性.采用离子交换柱和凝胶柱对FCA进行分离纯化得到分子量 28.188 kDa的多糖组分PFCA2,单糖组成结果表明,PFCA2主要由鼠李糖、葡萄糖、阿拉伯糖、半乳糖、葡萄糖醛酸和半乳糖醛酸组成.体外抗氧化试验表明,PFCA2具有良好的DPPH自由基和羟基自由基清除活性,其IC50 值分别为0.504 1和 3.280 2 mg/mL;FCA具有良好的抗氧化能力,是天然的抗氧化剂.该研究结果为无花果工业副产物精深加工提供理论依据.
Separation and Purification Process and Antioxidant Performance of Ficus carica L.Polysaccharides
To better utilize by-products of Ficus carica L.and improve their economic value,antioxidant properties of Ficus carica L.polysaccharides(FCW)were investigated.Microwave-assisted extraction of FCW was carried out,and extraction process was optim-ized.The optimal conditions were that,extraction temperature was 70 ℃,solid-liquid ratio was 1:40,microwave power was 800 W,and extraction time was 50 min.Under these conditions,extraction rate of FCW was 12.32%.The residue was used to simulate waste after Ficus carica L.production,Ficus carica L.residue polysaccharides(FCA)was extracted using alkaline extraction method,and analyze their thermal stability.Results showed that FCA had good thermal stability below 260 ℃.Polysaccharide component PFCA2 with a molecular weight of 28.188 kDa was obtained by separating and purifying it by ion exchange column and gel column.Results of monosaccharide composition showed that PFCA2 was mainly composed of rhammnose,glucose,arabinose,galactose,glucuronic acid,and galacturonic acid.In vitro antioxidant experiments showed that PFCA2 had good scavenging activities for DPPH free radicals and hy-droxyl free radicals,with IC50 values of 0.5 041 mg/mL and 3.2 802 mg/mL.This study showed that FCA have good antioxidant capa-city and were natural antioxidants.This study could provide a theoretical basis for deep processing of industrial by-products of Ficus carica L..
