Laboratory study on the effect of microfocused ultrasound on isolated animal skin
Objective To investigate the histopathological characteristics of microfocused ultrasound(MFUS)on isolated animal skin tissues and the distance from the surface of the skin so as to provide reference for the safe use of microfocused ultrasound equipment in clinic.Methods Porcine skin tissue was cut into tissue blocks of 60mm × 40mm × 10mm and parallel operation lines were marked.Different treatment heads of the Peninsula MFUS ultrasound therapeutic instrument were used to simulate the isolated porcine skin tissue along the labeling line.Subsequently,the porcine skin tissue at the marked lines was cut into 10mm x 5mm tissue blocks for embedding,fixation,sectioning,and HE staining.The HE staining results of porcine skin tissues were observed under microscope,and the distance between heat injury area and skin surface was measured.Results Under the action of MFUS M4.5 and M3.0,there were no significant pathological changes in the dermis and epidermis tissues of pig skin.However,in the subcutaneous adipose tissue,there were varying degrees of adipose cell dissolution and changes in the form of transparent lacunas.The minimum distance of transparent lacunas from the skin surface under M4.5 and M3.0 was 2.68mm and 2.99mm,respectively,and the maximum distance was 4.92mm and 4.91mm,respectively.HE staining of porcine skin treated with D4.5 and D3.0 showed that deep collagen degeneration could be seen in porcine dermis,and changes in solubility of adipocytes of different sizes could be seen in subcutaneous adipose tissue,forming a transparent cavity,which was long and oval in shape.The distance from the top and bottom of collagen denaturation areas in pig dermis to the skin surface under D4.5 and D3.0 was 0.41mm,2.58mm,2.58mm and 2.54mm,respectively.And the maximum distances of transparent lacunas from the skin surface were 4.70mm and 4.92mm,respectively.Conclusion MFUS can cause adipose dissolution and dermal collagen fiber denaturation in skin tissues.Therefore,it is necessary to grasp the indications during operation to avoid operating in sunken areas,and avoid excessive overlap to prevent epidermal damage when using D3.0 treatment head.
