Functional Study of mmu_circRNA_009396 in Cardiac Hypertrophy
To investigate the regulatory role of circRNA009396 in cardiac hypertrophy,angiotensin Ⅱ(AngⅡ)was used to construct the hypertrophy model of neonatal mouse ventricular cardiomyocytes(NMVCs)in vitro.Hypertrophy marker genes were detected by RT-qPCR,and myocardial cytoskeleton was staining with phalloidin.The results show that the expression levels of circRNA009396 are down-regulated after 1 000 nmol/L Ang Ⅱ stimulation for 24 h,and down-regulated in the heart tissue of mice injected with Ang Ⅱ.Knockdown of circRNA009396 increases Ang Ⅱ-induced cardiac hypertrophy,and overexpression of circRNA009396 inhibits Ang Ⅱ-induced cardiac hypertrophy.This suggests that circRNA009396 is a cardio-specific circular RNA with cardioprotective effects,and may act as a key regulator to co-regulate the process of myocardial hypertrophy with downstream signaling pathways.
