A study on rapid propagation of aquatic plant Rotala rotundifolia'Gontin'by tissue culture
Rotala rotundifolia'Gontin'is a kind of aquatic plants,which has high ornamental and eco-logical value,but the traditional propagation method is difficult to meet the market demand.In order to ob-tain a large number of R.rotundifolia'Gontin'seedlings in a short period of time,the buds were selected as explants,the optimum propagation method of plant tissue culture was studied using MS(Murashige and Skoog)as basic medium.The results showed that MS with 0.5 mg/L 6-BA(6-Benzylaminopurine)was the optimal medium for bud proliferation,with 21.1 buds and 9.83 mg/L chlorophyll content.Adding 1 g/L Activated carbon(AC)in MS medium was beneficial to rooting,and the optimum medium for rooting was MS with 1 g/L AC and 0.6 mg/L IBA(Indole-butyric acid).The transplant survival rate was 100%.In this research,a high frequency establishment of in vitro propagation system on R.rotundifolia'Gontin'will provide a reliable technical support for its large scale production and utilization.
