天祝白牦牛全线粒体基因组母系遗传多样性

为从基因组水平上揭示天祝白牦牛的母系遗传多样性、群体遗传结构和母系遗传背景,本研究利用基因组重测序技术,测定42头天祝白牦牛的全基因组序列,获取其线粒体基因组全序列,并从GenBank中下载1条已公布的天祝白牦牛线粒体基因组全序列,对共计43条天祝白牦牛线粒体基因组全序列进行综合分析.结果表明:从43条天祝白牦牛线粒体基因组全序列中共识别出971个变异位点,包括25个单一多态位点和946个简约信息位点;通过序列间核苷酸变异确定了 20种单倍型,单倍型多样度和核苷酸多样度分别为0.907和0.013,表明天祝白牦牛拥有较为丰富的母系遗传多样性.与野牦牛及环湖、雪多、玉树、青海高原、帕米尔等牦牛品种相比,天祝白牦牛母系遗传多样性相对较低.利用美洲野牛作为参考外群进行系统发育分析,20种单倍型分布于5种单倍型组(A、B、C、E和G)中,归属于3个母系遗传支系(Mt-Ⅰ、Mt-Ⅱ和Mt-Ⅲ),以Mt-Ⅰ支系为主,推测其有3个母系起源.
Maternal genetic diversity of complete mitogenome of Tianzhu white yak
To reveal the maternal genetic diversity,population genetic structure and maternal genetic background of Tianzhu white yak at genomic level,genome re-sequencing technology is adopted to determine the complete genome sequence of 42 Tianzhu white yaks to obtain their complete mitoge-nome sequences.Besides,one published complete mitogenome sequence of Tianzhu white yak is downloaded from GenBank.A comprehensive analysis is conducted on 43 complete mitogenome se-quences of Tianzhu white yaks.The results show that a total of 971 mutation sites are identified in 43 complete mitogenome sequences,including 25 single polymorphism sites and 946 parsimony informa-tive sites.A total of 20 haplotypes are identified through nucleotide variations between sequences,with haplotype diversity and nucleotide diversity of 0.907 and 0.013 respectively,indicating that Tianzhu white yak has relatively rich maternal genetic diversity.The maternal genetic diversity of Tianzhu white yak is relatively lower than that of wild yak,Huanhu,Xueduo,Yushu,Qinghai plateau and Pamir yaks.Taking Bison bison as a reference outgroup for phylogenetic analysis,it is found that 20 haplotypes are distributed in 5 haplotype groups(A,B,C,E and G),belonging to 3 maternal lineages(Mt-Ⅰ,Mt-Ⅱ and Mt-Ⅲ),with Mt-Ⅰ dominated.Therefore,it is speculated that there are 3 maternal origins.
