一株柚子叶部病原真菌的鉴定及生物学特性研究

分离鉴定柚子炭疽病的病原菌,对其致病性和生物学特性进行研究.利用组织分离法对柚子炭疽病的病原菌进行分离纯化;利用离体叶片接种方法检测其致病性;通过形态学观察和ITS基因序列同源性比对及其系统进化分析确定其分类地位;利用单因素试验研究其生物学特性.结果表明,分离得到的致病真菌YZ4被鉴定为胶孢炭疽菌(Colletotrichum gloeosporioides);最适合YZ4生长的碳源为淀粉,氮源为蛋白胨,最适生长pH为5.0.研究结果为柚子炭疽病的防治提供基础数据.
Identification and Biological Characteristics of a Pathogenic Fungi from the Leaves of Pomelo
To isolate and identify the pathogens resulting Citrus maxima anthracnose as well as study its pathogenicity and biological characteristics,tissues isolation was used to isolate and purify the pathogens of an-thracnose in C.maxima.In vitro leaf inoculation test was used to confirm the pathogenic of the isolated poten-tial pathogens,and its taxonomic status was determined by morphological observation,sequence homology com-parison and phylogenetic analysis of ITS gene.Biological characteristics of the fungal was determined by single factor test.The results indicated that the isolated C.maxima pathogenic fungi named YZ4 was identified as Colletotrichum gloeosporioides.The most suitable carbon and nitrogen sources for YZ4 growth were soluble starch and peptone,respectively.The optimal growth pH of the strain was 5.0.The above results provide basic data for the management of anthracnose in C.maxima.
