为了解白刺遗传水平的特征,本文以唐古特白刺叶片为材料,利用高通量测序技术进行转录组分析.基因注释结果显示,总共注释了 28 614个Unigenes,七大数据库共同注释到的有3 176个.NR注释结果表明唐古特白刺叶片基因序列与甜橙的同源序列匹配度最高.GO数据库基因注释主要集中在细胞过程、代谢过程和结合与催化活性.KEGG数据库中基因在细胞过程分支中富集到了共316个,环境信息处理分支中富集到了 250个,其中植物激素信号调节代谢途径在分支中注释到的基因最多,有223个,它在白刺干旱调节过程中发挥着重要作用,在新陈代谢分支中氨基酸代谢途径、苯丙烷类物质生物合成等途径中注释到的显著表达的基因较多,这与唐古特白刺的应激耐受性和其生境的恶劣环境有关.
Determine and Analysis of Leaf Transcriptome of Nitraria tangutorum
In order to understand the genetic characteristics of Nitraria tangutorum Bobr.at the genetic level,this paper took the leaves of Nitraria tangutorum Bobr.as materials for transcriptome analysis using high-throughput sequencing technology.The gene annotation results showed that a total of 28 614 Unigenes were annotated,and 3 176 were jointly annotated by seven major databases.The NR annotation results indica-ted that the gene sequence of Nitraria tangutorum Bobr.leaves had the highest homology sequence matching degree with sweet oranges.The gene annotation in the GO database was mainly concentrated in cellular processes,metabolic processes,and binding and catalytic activities.In the KEGG database,a total of 316 genes were enriched in the cellular process branch and 250 genes were enriched in the environmental informa-tion processing branch.Among them,the plant hormone signal-regulated metabolic pathway played an im-portant role in the drought regulation process.The branch with the most genes annotated was 223.In the me-tabolism branch,significantly expressed genes were more frequently annotated in metabolic pathways such as amino acid metabolism pathways and phenylpropanoid biosynthesis,which were related to the stress tolerance of Nitraria tangutorum Bobr.and the harsh environment of its habitat.
