Pathogen Isolation,Identification and Bioinformatics Analysis of Clostridium Septicum of Yak Origin
In order to study the biological characteristics of Clostridium septicum isolated from yaks in Guoluo area of Qinghai Province,three organ samples of dead yaks from Maqin County Veterinary Station in Guoluo Prefec-ture of Qinghai Province were isolated and cultured,identified and analyzed for biological characteristics,and the i-solated pathogenic strains were specifically amplified by PCR and compared with phylogenetic trees.The results showed that the strain isolated from the spleen of dead cattle was Gram-positive bacilli,which grew well in VF medium PH8.0 and could grow on PH8.0's sheep blood Agar medium with smooth protruding,round edges and hemolysis,and the pure culture was diluted step by step and subcutaneously injected into 0.2 mL Kunming mice,all the mice died within 24 hours.From the growth curve,it can be seen that 0~2 h is the slow phase,>2~14 h is the logarithmic phase,>14~24 h is the stable period,and begins to decline after 24 h.The specific am-plification results showed that the size of 1 300 bp was consistent with the expected results,and the target band of 1 450 bp was obtained by 16SrRNA amplification.The results of phylogenetic tree analysis showed that the isolated strain was Clostridium septicum.Through laboratory isolation and identification,combined with the results of mo-lecular biology experiments,it was diagnosed as yak Clostridium septicum,which provided technical support for the prevention and control of the disease in this area.
