四妙散及其拆方对类风湿关节炎大鼠血清IL-6、MMP-3的影响

目的:观察四妙散及其拆方对经典RA动物模型血清IL-6、MMP-3 的影响,以了解四妙散对RA的治疗效果及其组方特点.方法:将 100 只雌性SD大鼠随机分成 10 组,随机分为正常对照组、模型对照组、西药对照组(甲氨蝶呤,MTX)、四妙汤全方组及其6 个拆方组(黄柏+牛膝、苍术+牛膝、薏苡仁+牛膝、苍术+薏苡仁、黄柏+薏苡仁、黄柏+苍术),每组 10 只.除空白组外,余组大鼠以每只0.1 ml弗氏完全佐剂于右侧足跖部注射诱导关节炎模型.造模后第 1 天开始药物干预,正常对照组和模型组分别给予等体积双蒸水灌胃,所有大鼠按照 10 ml/kg 灌胃给药,1 次/d,连续灌胃 14 d.灌胃治疗结束后,采用ELISA法测定各组大鼠血清中IL-6、MMP-3 的含量.结果:四妙散全方及拆方组大鼠血清IL-6、MMP-3 均有不同程度下降(P<0.05).根据综合指标结果,四妙散全方效果最佳,其次分别为黄柏+牛膝组、黄柏+苍术组、苍术+牛膝组、黄柏+薏苡仁组、薏苡仁+牛膝组、苍术+薏苡仁组.结论:四妙散全方的配伍应用,发挥了降低佐剂诱导致RA大鼠血清炎症因子最佳作用效果,拆方两两组合效果最强为黄柏+牛膝组.四妙散全方组对降低佐剂诱导致RA大鼠血清炎症因子效果明显,体现了四妙散组方药简力专的鲜明特点.两两组合中黄柏+牛膝组对大鼠血清IL-6、MMP-3 含量下降效果最强,对四妙散的君臣配伍有了新的认识,但尚需进一步临床研究证实.
Effects of Simiao Powder and Its Disassembled Prescription on Serum IL-6 and MMP-3 in Rats with Rheumatoid Arthritis
Objective:To observe the effects of the classic RA animal model serum IL-6 and MMP-3 with the "Simiao Powder"and its disassembled formulas,in order to understand the therapeutic efficacy of"Simiao Powder"for RA and its composition characteristics.Methods:100 female SD rats were randomly divided into 10 groups,including a control group,a model group,a western medicine group(methotrexate,MTX),a"Simiao Decoction"group,and six disassembled formula groups(Phellodendron+Achyranthes,Atractylodes+Achyranthes,Coix Seed+Achyranthes,Atractylodes+Coix Seed,Phellodendron+Coix Seed,and Phellodendron+Atractylodes).Each group had 10 rats.Except for the blank group,the remaining rats were induced with arthritis models by injecting 0.1 ml of Freund's complete adjuvant on the right foot.Starting from the first day after modeling,drug intervention began in each group.The normal control group and model group were given equal volumes of double-distilled water by gavage.All rats were administered according to a dosage of 10 ml/kg once daily for 14 consecutive days.After the gavage treatment was completed,the levels of IL-6 and MMP-3 in the serum of each group were measured using the ELISA method.Results:The levels of IL-6 and MMP-3 in the serum of rats in the "Simiao Powder"group and its disassembled formulas decreased to varying degrees(P<0.05).According to the comprehensive index results,the "Simiao Powder"group had the best effect,followed by the Phellodendron+Achyranthes group,Phellodendron+Atractylodes group,At-ractylodes+Achyranthes group,Phellodendron+Coix Seed group,Coix Seed+Achyranthes group,and At-ractylodes+Coix Seed group.Conclusion:The combined application of the "Simiao Powder"has exhibited optimal efficacy in reducing serum inflammatory factors in adjuvant-induced RA rats.The strongest effect is observed in the Phellodendron+Achyranthes group.The "Simiao Powder"group has a notable characteristic of concise and specialized composition.Among the pairwise combinations,the Phellodendron+Achyranthes group has the strongest effect in reducing serum IL-6 and MMP-3 levels in rats.This provides new insights into the dominant and supporting components of"Simiao Powder",but further clinical studies are needed to confirm these findings.
