Experimental Study on Cistanche deserticola Bone Targeting Nanoparticles Promoting Bone Defect Repair in Rats
Objective:To observe the effect of Cistanche deserticola bone targeted nanoparticles on pro-moting the repair of bone defects in rats. Methods:Forty clean male SD rats were randomly divided into four groups using a random number table method, namely a normal control group, tetracycline nanoparticles(TC-PLGANP)group,Cistanche deserticola nanoparticles(CH/PLGANP)group,and Cistanche deserticola bone targeted nanoparticles(CH/TC-PLGANP)group,with 10 rats in each group.Except for the normal control group, each group was established using a corresponding method to establish a rat femoral defect model. After drug intervention, western blot and immunohistochemical methods were used to detect the expression levels of FGF-2,VEGF,and BMP-2 in bone tissues. Results:Compared with the normal control group, the relative expression of FGF-2,VEGF,and BMP-2 proteins and the relative expression of positive particles in bone tis-sues of rats in the TC-PLGANP group,CH/PLGANP group, and CH/TC-PLGANP group significantly in-creased(P<0.05).Compared with the TC-PLGANP group, the relative expression of FGF-2,VEGF,and BMP-2 proteins and the relative expression of positive particles in bone tissues of rats in the CH/PLGANP group and CH/TC-PLGANP group significantly increased(P<0.05).Compared with the CH/PLGANP group, the relative expression of FGF-2,VEGF,and BMP-2 proteins and the relative expression of positive particles in bone tissues of rats in the CH/TC-PLGANP group significantly increased(P<0.05).Conclu-sion:Compared with ordinary preparations of Cistanche deserticola, bone targeted preparations of Cistanche de-serticola have higher expression levels of bone repair factors in bone tissues, which is beneficial to the repair of bone defects.
