Effect of alcohol extract of shui medicine Gnetem parvifolium on serum IL-6 and TNF-ɑ levels and the expression of VEGF and MMP-3 in soft tissue of ankle joint in rats with rheumatoid arthritis
Objective:To observe the effects of the alcohol extract of Gnetem parvifolium on serum levels of TNF-ɑ and IL-6,as well as the expression of VEGF and MMP-3 in the soft tissue of the ankle joint in rats with type II collagen-induced arthritis.This study aims to preliminarily explore the pharmacological mecha-nism of Gnetem parvifolium in treating rheumatoid arthritis.Methods:Using a rat model induced by type II col-lagen,60 SD rats were randomly divided into a blank group,a model group,and three groups of low,medium,and high doses of the alcohol extract of Gnetem parvifolium,with 10 rats in each group.After initial inflamma-tion for 24 hours,except for the blank and model groups,rats were administered orally once daily for four weeks.The levels of IL-6 and TNF-ɑ were measured using ELISA,and the expression of VEGF and MMP-3 in the soft tissue of the ankle joint was determined using immunohistochemical methods.Results:Compared to the blank group,the levels of IL-6 and TNF-ɑ in serum and the expression of VEGF and MMP-3 in the soft tissue of the ankle joint in the model group significantly increased(P<0.05).Compared to the model group,the levels of IL-6 and TNF-ɑ in serum and the expression of VEGF in the soft tissue of the ankle joint in rats from the low,medium,and high dose groups significantly decreased(P<0.05),and the expression of MMP-3 in the soft tissue of the ankle joint in rats from the medium and high dose groups significantly decreased(P<0.05).Conclusion:The alcohol extract of Gnetem parvifolium can reduce serum levels of IL-6 and TNF-ɑ as well as the expression of VEGF and MMP-3 in the soft tissue of the ankle joint in rats with rheumatoid ar-thritis.This suggests that it can inhibit the release of inflammatory factors and alleviate the progression of RA.
