Extraction and anti-oxidant activity of Cannabis polysaccharide
Cannabis polysaccharide-I was obtained by water extraction and alcohol precipitation from the residue of cannabis soaked in ethanol.Polysaccharide-Ⅰ was obtained from the residue through water extraction and alcohol precipitation.The water phase was concentrated and then extracted with ethyl acetate,resulting in the isolation of cannabis polysaccharide-Ⅱ.After ethyl acetate extraction,the aqueous layer was further processed with n-butanol and concentrated under reduced pressure to yield cannabis polysaccharide-Ⅲ.The polysaccharide content of the three types of cannabis polysaccharides was determined using the phenol-sulfuric acid method.Additionally,scavenging experiments were conducted to evaluate their antioxidant activity against DPPH,ABTS,hydroxyl,and superoxide anion free radicals.The experimental results demonstrated that the three types of cannabis polysaccharides had good effects on DPPH clearance rate and ABTS clearance rate.Among them,polysaccharide-Ⅱ showed the strongest antioxidant activity,with IC50 values for scavenging DPPH,ABTS,hydroxyl radical,and superoxide anion radicals at 0.012,0.043,0.138,1.857 mg/mL,respectively.
