火焰兰等5种兰花菌根显微结构及分离培养基筛选

通过石蜡切片法对5种不同类型的兰花新鲜营养根的显微结构进行研究,并利用组织分离法对其中的内生真菌进行分离,从而筛选最佳的分离培养基.结果表明:5种兰花菌根结构一致,由外到内依次为根被、皮层、中柱,三个部位都有不同程度的被真菌侵染表现,海南钻喙兰、火焰兰是通过皮层细胞直接侵染,卷萼兜兰、硬叶兜兰和铁皮石斛是通过根毛或皮层细胞侵染,并形成菌丝团.实验结果显示:PDA培养基最适合做5种兰花菌根分离纯化培养基.
Endomycorrhizal Structure and Solid Medium Selection of Five Orchid Species including Renanthera coccinea Lour
Using five orchid species as experimental materials,Observing the microstructure of mycorrhizal fungi using paraffin sectioning method.And using tissue isolation method to isolate and culture endophytic fun-gi,for selecting the optimal culture medium.The results showed that the root structures of the five orchids are root cover,cortex and stele which all infected with mycorrhizal fungi to varying degrees.Epiphytic orchids are directly infected by cortical cells.Saprophytic orchids are infected by root hairs or cortical cells,and form a mycelium mass.PDA medium is the most suitable medium for isolation and purification of orchid mycorrhizal fungi.
