Identification,Biological Characterization,and Fungicide Screening of Pathogens Causing Anthracnose in Jasminum sambac
Biological characterization and affective fungicide screening studies were performed to clarify the pathogenic bacterial species of jasmine leaf anthracnose disease in Hengzhou city,Guangxi Province,China.Through the isolation and purification of the pathogen,identification of pathogenicity and morphological observation,a phylogenetic tree,based on the sequencing results of ITS,ACT,CHS-1,TUB2,GAPDH,and ApMat,was constructed for the representative strain HL6-1 by using MEGA v.10.1.5 software,and it was clarified that the strain was Colletotrichum siamense,which is an anthracnose fun-gus of Siam.The aerial mycelium of the strain was fluffy,and the colonies were initially white and then became dark gray with the production of an orange spore mass;the conidia were colorless,had an overall long ellipsoid shape,and were single-celled,with an average size of 14.58 μm×5.97 μm.The results of biological characterization showed that the optimal cultivation temperature of this strain was 28℃,the optimal pH was 7,the optimal carbon source was glucose,and the optimal nitrogen source was sodium nitrate.Finally,the mycelial growth rate method was used for indoor virulence determination to screen for effective fungicides,and the results showed that pyraclostrobin,phenyl ether metronidazole,and imidacloprid were more ef-fective at inhibiting the fungus,with EC50 values less than 1 mg·L-1.
