Extraction and Identification of Total Flavonoids from Pericarp of Seedless Wampee Fruit and Analysis of Their Antioxidant Activity
To extract high-purity total flavonoids from the pericarp of seedless wampee,the common flavonoid components were qualitatively and quantitatively analyzed,and the in vitro antioxidant activities of the purified total flavonoids were in-vestigated.Ultrasonic-assisted alcohol extraction and macroporous resin purification methods were used to get total flavonoid extract from dried pericarp,and the main components and contents of flavonoids were detected by UPLC-HRMS(ul-tra-performance liquid chromatography-high-resolution mass spectrometry)system,and in vitro antioxidant ability was tested via physiological and biochemical methods.The results indicated that the maximum yield of total flavonoids could be obtained by ultrasonic extraction for 60 min and twice with 75%ethanol at a 1:14(g.mL‒1)solid-liquid ratio.The total flavonoid con-centration in the extract reached 43.2 mg/mL and increased to 235 mg/g with a purity of 86.45%after purification.LC-MS/MS detection results revealed that rutin,quercetin,kaempferol,and isorhamnetin were the main components,and their contents ranged from high to low.When the total flavonoid extract concentration was higher than 1 mg.mL‒1,the total antioxidant and free radical scavenging capacities were positively correlated with the flavonoid concentration.
