Optimization of Traditional Seedling Breeding Methods for Dendrobium officinale via Orthogonal Experiments
To improve the germination rate of axillary buds in traditional Dendrobium officinale seedlings and guide the promotion and application of traditional seedling cultivation techniques in production.,a study was conducted on the factors influencing germination and seedling rates of traditional seedling-raising axillary buds.A four-factor and five-level orthogonal experiment was designed for the breeding mode(A),hormone treatment(B),seedling substrate(C),and strains(D).The results revealed that the prime and secondary order of factors affecting the germination and seedling rate of axillary buds in traditional seedling cultivation was D>A>B>C.The optimal levels of the four factors are A2(the whole strip is laid flat and covered with a substrate for approximately 1 cm),B2(NAA 0.5 mg·L-1+6-BA 0.5 mg·L-1),C1(fresh moss),D4(purple sprite),and the optimal combination of traditional seedling cultivation factors A2B2C1D4 was selected through experiments,and its germination and seedling rates can reach 81%,which is significantly higher than the original natural axillary bud germination and seedling rates of 10%~20%.
