Effect of Seeding Macrotyloma uniflorum and Vicia sativa Plants with Different Agents on Seed Germination under PEG-6000 Stress
This study aimed to identify optimal seed-dipping drugs and concentrations to promote the germination of M.uniflorum and V.sativa under drought stress.Seeds of M.uniflorum and V.sativa were used as test materials.Drought treat-ment was simulated with concentrations of 2%,5%,and 10%polyethylene glycol 6000(PEG-6000),and the seeds were treated with chitosan,salicylic acid,calcium chloride,and vinblastine.The seeds were soaked in distilled water as a control,and the germination rate,germination index,root fresh weight,root length,root volume,and root surface area were measured.The results of the study revealed that the germination of M.uniflorum and V.sativa was more than that at 2%PEG-6000,and the germination of M.uniflorum and V.sativa seeds was inhibited at 5%and 10%PEG-6000.The measured indices of V.sa-tiva treated with drugs in a 2%PEG-6000 solution decreased with increasing drug concentration.The germination indices de-creased with increasing concentrations of chitosan,salicylic acid,and calcium chloride,and the highest values were reached at concentrations of 50,50,500,and 700 mg·L-1 respectively.M.uniflorum peas did not germinate in the 50,200 mg·L-1 salicylic acid and 1,000 mg·L-1 calcium chloride treatments and tended to decrease but then increased in the chitosan and vin-blastine treatments.The six indices of the germination process of the green manure seeds were evaluated comprehensively according to their affiliated functions.Drug triggering could alleviate the effects of slight drought stress on seed germination,and drug-impregnated seeds were used to promote germination in the M.uniflorum and V.sativa seeds.The best treatments for promoting the germination of M.uniflorum and V.sativa seeds at 24 h were 1 400 mg·L-1 ethylene glycol solution and 50 mg·L-1 chitosan solution,respectively.
