基于Sanger测序技术的云南荔枝园蠹虫种类初步鉴定

荔枝作为云南乡村振兴重要产业之一,随着面积的不断扩大,其种植生态环境多样化,病虫害的种类也随之增加.近几年在云南荔枝主要产区陆续发现蠹虫为害,调查荔枝蠹虫为害情况并鉴定其种类,对云南荔枝病虫害防治具有重要意义.采用 5 点取样法,针对云南荔枝 5 个主要产区蠹虫为害程度以及为害状进行调查,利用Sanger测序技术进行生理小种初步鉴定.结果发现,5 个产区均有不同程度蠹虫为害,为害状与长蠹科昆虫为害状相似.经 NCBI数据库比对,初步将本次调查的蠹虫鉴定为 2 属 3 种,分别是双棘长蠹属侧突双棘长蠹(Sinotylon perforans)、双棘长蠹(Sinoxylon anale)和长蠹属槲长蠹(Bostrichus capucinus).
Preliminary Identification of Beetle Species in Litchi Orchards in Yunnan Based on Sanger Sequencing Technology
As one of the crucial industries for rural revitalization in Yunnan Province,litchi has diversified its planting eco-logical environment with the continuous expansion of its area,and the types of diseases and pests have also increased.Beetles have been discovered in the main production areas of litchis in Yunnan in recent years.Investigating the harm caused by litchi beetles and identifying their types is highly important for the prevention and control of lychee diseases and pests in Yunnan.This study adopted a 5-point sampling method.First,a survey was conducted on the degree and status of damage caused by beetles in the five main production areas of litchi in Yunnan.Sanger sequencing technology was used for preliminary identifi-cation of physiological races.There were varying degrees of damage caused by beetles in all five production areas,and the damage pattern was similar to that of insects in the family Scorpaenidae.After Sanger sequencing technology was used for detection,a preliminary identified through comparison in the NCBI database revealed that there are two genera and three spe-cies,namely Sinotylon perferans,Sinoxylon manage,and Bostrichus capuchinus.
