The Mechanism of the Treatment of Diabetic Nephropathy with Breviscapus was Studied Based on Network Pharmacology
To study the mechanism of Chinese herbal medicine Calendula breviscapus in the diabetic nephropathy(DN)treatment via network pharmacology,through the TCMSP platform,ETCM database,and PubChem database,the active in-gredients and targets of Erigeron brevis were screened.Target genes related to DN were obtained via DisGenet and GeneCards.To screen the common target genes of the active ingredients of Erigeron breviscapus and the disease targets,we constructed a"component-target-disease"network diagram via Cytoscape 3.9.1 software.The protein interaction(PPI)network map was built via the STRING online database.KEGG and GO analyses of common targets were performed via the Cytoscape 3.9.1 software plug-in ClueGo to screen potential signaling pathways and analyze their mechanisms of action.Nine active ingredients of E.breviscapus were obtained through screening,and 79 targets of its action on DN were identified.GO en-richment analysis revealed that the biological functions of the genes associated with the interaction between breviscapus and diabetic nephropathy were related mainly to the positive regulation of the transmembrane receptor protein tyrosine kinase signaling pathway and the negative regulation of cell proliferation and apoptosis.The enrichment results of the KEGG path-way analysis revealed that the intersecting genes were distributed mainly in the PI3K-AKT signaling pathway.The Chinese herbal medicine E.breviscapus is a species of the Compositae family.It has the functions of dispelling wind and dampness,dispelling cold and dissolving surfaces,clearing collars,and relieving pain.Its mechanism of action shows the characteristics of multi-component,multi-target,and multi-pathway,and it is often used as a drug to prevent and treat various diseases in the clinic.This study investigated the mechanism of action of E.breviscapus in the treatment of DN from two aspects of the pharmacological action and network pharmacology,which is conducive to a more in-depth and comprehensive understanding of the medicinal value of E.breviscapus,and it provides a theoretical reference for its development and utilization.
