The expression of GP73 and miR-218-5p in human colorectal cancer and their relationship with clinicopathological features and prognosis of patients
Objective To investigate the expression of Golgi protein 73(GP73)and miR-218-5p in patients with colorectal cancer and their relationship with clinicopathological features and prognosis.Methods A total of 100 patients with colorectal cancer who were treated in the Zhangye People's Hospital Affiliated to Hexi University from March 2018 to March 2019 were selected as the study group,and 61 healthy volunteers who underwent physical examination in the hospital during the same period were selected as the control group.The expression differences of GP73 and miR-218-5p between the two groups were compared.The expression of GP73 and miR-218-5p in patients with different pathological characteristics of colorectal cancer was compared.The relationship between GP73 and miR-218-5p and the prognosis of patients with colorectal cancer was analyzed by Kaplan-Meier curve.Multivariate Cox proportional hazard model was used to analyze the factors affecting the prognosis of patients with colorectal cancer.Results The level of GP73 in the study group was significantly higher than that in the control group,and the level of miR-218-5p was lower than that in the control group(t=33.030,30.578;all P<0.05).The levels of GP73 and miR-218-5p were correlated with different TNM stages and lymph node metastasis(t=3.904,11.149;all P<0.05).Kaplan-Meier curve analysis showed that the survival rate of GP73 high expression group was significantly lower than that of GP73 low expression group(x2=19.785,P<0.05),and the survival rate of miR-218-5p high expression group was significantly higher than that of miR-218-5p low expression group(x2=8.827,P<0.05).Multivariate Cox analysis showed that TNM stage Ⅲ+Ⅳ(HR=2.187),lymph node metastasis(HR=1.065),high expression of GP73(HR=3.182)and low expression of miR-218-5(HR=3.613)were the influencing factors of prognosis in patients with colorectal cancer(P<0.05).Conclusions The level of GP73 in patients with colorectal cancer was significantly increased,and the level of miR-218-5p was significantly decreased.The abnormal expression of GP73 and miR-218-5p was related to TNM stage and lymph node metastasis.Colorectal patients with high expression of GP73 and low expression of miR-218-5p had poor prognosis.
