Expression and diagnostic value of serum miR-6867-5p in patients with Alzheimer's disease
Objective To investigate the expression level and clinical diagnostic value of serum miR-6867-5p in patients with Alzheimer's disease(AD),and analyze the possible mechanism of miR-6867-5p target gene involved in AD.Methods Selected 52 patients with AD who visited the neurology clinic of the Affiliated Hospital of Jiangsu University from November 2021 to June 2022 as the AD group,and 28 persons as the control group,who underwent routine physical examination in the physical examination center of the hospital during the same period.Blood samples and basic clinical data were collected from the two groups.The expression level of miR-6867-5p was detected by real-time fluorescence quantitative PCR before and 6 months after donepezil treatment in AD group.The cognitive function of AD patients was evaluated by minimum mental state examination(MMSE)and Montreal cognitive assessment(MoCA).The correlation between the expression level of serum miR-6867-5p and the scores of MMSE and MoCA were analyzed,and receiver operating characteristic(ROC)curve was used to assess the diagnostic value of miR-6867-5p in AD.The online prediction tools were used to predict the target genes of miR-6867-5p.Functional enrichment analysis of the miRNA target genes was performed by the R language analysis package.Results The expression level of serum miR-6867-5p in AD group before treatment was higher than that in control group(Z=-4.65),and the expression level of serum miR-6867-5p after treatment was lower than that of before treatment(Z=-5.54),with statistically significant differences(all P<0.01).There was significant negative correlation between expression level of serum miR-6867-5p and MMSE,MoCA scores(r=-0.72,-0.69;all P<0.01).ROC curve showed that the area under the curve of serum miR-6867-5p was 0.82(95%C/:0.73-0.91,P<0.01),and the sensitivity was 80.77%,and the specificity was 71.43%.The online prediction tool obtained a total of 1 277 identical target genes.The results of gene ontology(GO)enrichment analysis showed that the target genes of miR-6867-5p were found in the synapse,and were mainly involved in axon development,modulation of chemical synaptic transmission regulation of trans-synaptic signaling and learning or memory.The molecular functions included DNA-binding transcription and so on.The results of Kyoto encyclopedia of genes and genomes(KEGG)analysis showed that miR-6867-5p was associated with glutamatergic synapse and Wnt signaling pathway.Conclusion The expression of serum miR-6867-5p was up-regulated in AD patients,and was expected to become a potential biomarker for clinical auxiliary diagnosis of AD.
