Effect of Rebamipide on the polarized phenotype of peritoneal macrophages in MSU induced peritonitis
Objective To observe the effect of Rebamipide on the polarized phenotype and cytokine expression of peritoneal macrophages in mice with acute gout peritonitis induced by monosodium urate(MSU).Methods Eighteen male BALB/c mice were randomly divided into three groups:MSU group(0.2 mL/10 g saline by gavage),Colchicum group(0.75 mg/kg colchicum 0.2 mL/10 g by gavage)and Rebamipide group(150 mg/kg rebamipide 0.2 mL/10 g by gavage),with 6 mice in each group.After two gavages,all three groups of mice were intraperitoneally injected with MSU once to prepare the acute peritonitis model.Sixteen hours after intraperitoneal injection of MSU,peritoneal macrophages were collected and their phenotypes were analyzed.Tumor necrosis factor-α(TNF-α),interleukin-6(IL-6)and IL-10 cytokines were detected in the supernatant of peritoneal lavage solution.Peritoneal tissue was stained with hematoxylin-eosin to analyze the inflammatory cell infiltration.Results The proportion of M2 type marker CD206 positive cells in the Colchicum group and Rebamipide group were(19.36±2.80)%and(21.22±1.22)%,respectively,which were significantly higher than those in the MSU group[(15.80±1.32)%],and the differences were statistically significant(both P<0.05).The concentrations of pro-inflammatory factors TNF-α and IL-6 in the peritoneal lavage fluid of Colchicum group and Rebamipide group were(138.73± 14.06),(122.89±6.66)μg/mL and(119.91±4.99),(115.32±6.73)μg/mL,respectively,and were significantly lower than those of the MSU group[(238.89±16.16),(272.99±11.35)μg/mL],the differences were statistically significant(all P<0.05);the concentrations of anti-inflammatory factor IL-10 in Colchicum group and Rebamipide group were(73.11±4.77),(70.69±3.72)μg/mL,respectively,which were significantly higher than those in the MSU group[(32.23±3.51)μg/mL)],the differences were statistically significant(all P<0.05).The HE staining results of mouse peritoneal tissue showed that the MSU group had loose peritoneal tissue,disordered cell arrangement,and a large number of inflammatory cells(with large nuclei,deep staining,and no cytoplasm)infiltration;The cells in Colchicum group and Rebamipide group showed edema and less infiltration of inflammatory cells compared to the MSU group.Conclusion Rebamipide mitigated MSU-induced inflammatory response in mice with gout peritonitis by increasing the M2 polarization of peritoneal macrophages.
