Phylogenetic analysis and population genetics of the Pomacea snail(Gastropoda:Ampullariidae)in Hainan province,China
Objective Molecular identification of the Pomacea snails in Hainan province was carried out to complete the study of genetic diversity and genetic relationships,which could provide a basis for identifying and preventing the continued spread of the Pomacea snails.Methods Based on mitochondrial cytochrome c oxidase I(CO1)and 16S ribosomal deoxyribonucleic acid(16S rDNA)genes,phylogenetic analysis and population genetics studies were conducted on Pomacea snail samples collected from 61 sampling points in 18 counties and cities in Hainan province.From the collected 1 050 Pomacea snails,632 and 577 of them were sequenced for COI and 16S rDNA genes.The relevant sequences were downloaded from the GenBank database to construct the database.The COI and 16S rDNA databases had 1 465 and 641 sequences respectively.Phylogenetic and genetic diversity analyzes were performed using MEGA X and DnaSP software.Results Pomacea canaliculata was found to be a dominant species(79.9%).The sympatric occurrence of P.canaliculata and P.occulta in 17 cities and counties was recorded.P.canaliculata isolates were categorized(based on COI)into clade A(closely related to strains from Japan and China),B(displaying proximity to strains from Japan,the Philippines,Malaysia and China),and C(closely related to strains from Malaysia,Japan,Argentina,Papua New Guinea,Myanmar,Chile and China).P.canaliculata was classified(based on 16S rDNA)into clade A and B(closely related to strains from Malaysia,Argentina,the Philippines,the USA and China).The haplotype network analysis revealed that P.canaliculata could be grouped into four and three haplotypes,corresponding to COI and 16S rDNA gene assemblages,respectively.Conversely,P.occulta exhibited only a single COI haplotype.Moreover,Pomacea specimens from Baoting Li and Miao Autonomous County had the highest haplotype and nucleotide diversities(Hd=0.679,π=0.053),whereas differentiation of the largest pairwise population existed between Qionghai city and Tunchang county(Fsr=0.63).The two Pomacea species had low genetic diversity.Conclusions The COI and 16S rDNA sequences of P.canaliculata exhibited the polymorphism.Apple snails might have been introduced to Hainan multiple times.However,P.occulta was confirmed for the first time,and no P.maculata had been recorded.
