Expression and prognostic value of SPP1 and miR-340-5p in thyroid cancer
Objective To investigate the expression of secreted phosphoprotein 1(SPP1)and microRNA-340-5p(miR-340-5p)in thyroid cancer and their relationship with prognosis.Methods From March 2018 to May 2020,119 patients with differentiated thyroid cancer(DTC)admitted to the Breast Surgery Department of The Fifth Clinical Medical College of Shanxi Medical University were selected as the study group;according to the recurrence,metastasis,and mortality during the follow-up period,they were grouped into a good prognosis group(n=84)and a poor prognosis group(n=35);another 119 patients with benign thyroid lesions who matched the general data with the study group were selected as the reference group.Pearson method was applied to analyze the correlation between serum SPP1 and miR-340-5p expression levels;multivariate logistic regression was applied to analyze the relevant factors affecting the prognosis of DTC patients;receiver operating characteristic(ROC)curve was applied to analyze the predictive efficacy of serum SPP1 and miR-340-5p on the prognosis of DTC patients.Results The expression levels of serum SPP1 and miR-340-5p in the study group were higher than those in the control group,the differences were statistically significant(t=15.732,10.111;all P<0.001);the expression levels of serum SPP1 and miR-340-5p in DTC patients were positively correlated(r=0.584,P<0.05);the expression levels of serum SPP1 and miR-340-5p in the poor prognosis group were higher than those in the good prognosis group,the differences were statistically significant(t=7.497,4.938;all P<0.001);the proportions of patients with lymph node metastasis and TNM stage Ⅲ+Ⅳ in the poor prognosis group were greatly higher than those in the good prognosis group,the differences were statistically significant(x2=11.297,6.802;all P<0.05);lymph node metastasis(OR=1.901),TNM stage Ⅲ+Ⅳ(OR=2.178),SPP1(OR=1.541),and miR-340-5p(OR=2.104)were the risk factors for poor prognosis in DTC patients(P<0.05);the AUC of serum SPP1 and miR-340-5p in predicting the prognosis of DTC patients was 0.829(95%CI:0.749-0.892)and 0.698(95%CI:0.607-0.779),respectively,while the AUC predicted by the combination of the two was 0.908(95%CI:0.841-0.953);the combination of the two was superior to the detection of serum SPP1 and miR-340-5p separately(Z=2.410,3.566;all P<0.05).Conclusion The expression levels of serum SPP1 and miR-340-5p were related to the occurrence of thyroid cancer,and the combined detection of the two had high predictive power for the prognosis of thyroid cancer patients.
