Methylation changes of PAX1 and TP63 gene promoter and their diagnostic value for CIN2+lesions in patients with cervical HPV infection
Objective To analyze the diagnostic value of pairedboxed gene1(PAX1)and TP63 gene promoter methylation for cervical intraepithelial neoplasia(CIN)2+lesions in patients with cervical human papilloma virus(HPV)infection.Methods The specimens of cervical tissue cells were collected from 116 patients with suspected cervical lesions from the Jingmen First People's Hospital between January 2016 and December 2022.According to the results of pathological examination,they were divided into control group(normal/cervicitis),low-grade squamous intraepithelial lesion(LSIL,including CIN1)group,high-grade squamous intraepithelial lesion(HSIL,including CIN2,CIN3)group and cervical cancer(CC)group.The methylation degree of PAX1 and TP63 gene promoters was detected by bisulfite sequencing method.The mRNA expressions of PAX1 and TP63 in cervical tissues were detected by real-time quantitative PCR(qPCR).DNA load of HR-HPV in cervical tissues was detected by second-generation hybrid capture method.The relationship between methylation of PAX1 and TP63 promoters and HPV infection was analyzed.HSIL and CC in pathological examination were classified as CIN2+,and the diagnostic efficiency of PAX1 and TP63 methylation detection for CIN2+was evaluated.Results The methylation levels of PAX1 and TP63 gene promoters were the highest in CC group,followed by HSIL group,LSIL group and control group,the differences were statistically significant(F=16.259,15.703;all P<0.05),while mRNA levels of PAX1 and TP63 were on the contrary,the differences were statistically significant(F=128.366,65.207;all P<0.05).DNA load of HR-HPV was the highest in CC group,followed by HSIL group,LSIL group and control group,the difference was statistically significant(F=92.514,P<0.05).Area under the curve(AUC)and cut-off values of PAX1 and TP63 methylation in the diagnosis of CIN2+lesions were(0.715,34.31 ct)and(0.694,35.28 ct),respectively.AUC of combined detection was 0.797,greater than that of single index(P<0.05).Conclusion The methylation degree of PAX1 and TP63 gene promoters could reflect the severity of cervical lesions,and their combined detection had high diagnostic value for CIN2+lesions.
