Expression and clinical significance of JAK2/STAT3 signaling pathway in peripheral blood of KOA patients with osteoporosis
Objective To analyze the expression of janus kinase 2/signal transducers and activators of transcription 3(JAK2/STAT3)signaling pathway in peripheral blood of patients with knee osteoarthritis(KOA)complicated with osteoporosis(OP)and its clinical significance.Methods KOA patients with OP who were treated in the Department of Orthopedics,Nanyang Central Hospital from January 2021 to December 2022 were selected as the study subjects and named as the combined group(n=106),and KOA patients diagnosed during the same period were selected as the control group(n=106).Age,sex,body mass index(BMI)and other general data of the two groups were collected,and JAK2,phosphorylation JAK2(p-JAK2),STAT3,phosphorylation STAT3(p-STAT3)protein expression levels were detected.Univariate analysis and multiple logistic regression were used to analyze the relationship between JAK2/STAT3 signaling pathway expression and KOA combined with OP in peripheral blood.The predictive value of JAK2/STAT3 signaling pathway expression in peripheral blood for KOA combined with OP was analyzed by receiver operating characteristic(ROC)curve.Results Univariate analysis showed that gender,age,BMI,housing condition,JAK2,p-JAK2,STAT3 and p-STAT3 protein expression were the influencing factors of KOA and OP,and the differences were statistically significant(c2/t=10.930,16.652,9.468,6.861,13.621,12.089,12.850,11.065;all P<0.05).Multivariate logistic regression analysis showed that gender(OR=1.397,95%CI:1.091-1.788),age(OR=1.631,95%CI:1.162-2.289),BMI(OR=2.210,95%CI:1.167-4.187),housing situation(OR=1.642,95%CI:1.082-2.493),JAK2 protein(OR=2.125,95%CI:1.257-3.594),p-JAK2 protein(OR=2.111,95%CI:1.285-3.466),STAT3 protein(OR=2.088,95%CI:1.320-3.302)and p-STAT3 protein(OR=2.140,95%CI:1.244-3.684)were independent risk factors for KOA combined with OP(all P<0.05).ROC curve analysis showed that the area under the curve(AUC)of JAK2,p-JAK2,STAT3,p-STAT3 and the combined detection of the four proteins were 0.898,0.880,0.897,0.857 and 0.985,respectively,and the combined detection of the four proteins was better than the single detection,the difference was statistically significant(P<0.05).Conclusions JAK2,p-JAK2,STAT3 and p-STAT3 proteins in peripheral blood of patients with knee osteoarthritis complicated with osteoporosis were highly expressed.KOA complicated with osteoporosis might be related to the active JAK2/STAT3 signaling pathway in peripheral blood.Combined detection had good predictive value for KOA complicated with osteoporosis.
