Expression levels and clinical significance of lncRNA XIST and miR-124 in peripheral blood monocytes during acute exacerbation of chronic obstructive pulmonary disease
Objective To explore the expression levels and clinical significant of long non-coding RNA X inactive specific transcript(lncRNA XIST)and miR-124 in peripheral blood mononuclear cells during acute exacerbation of chronic obstructive pulmonary disease(COPD).Methods A total of 123 COPD patients in the acute stage treated in the First Hospital of Handan from November 2020 to March 2023 were selected as the acute stage group;121 COPD patients in the stable stage group,and 122 healthy volunteers who underwent physical examination in our hospital during the same period were selected as the control group.The expression levels of interleukin-1β(IL-1β),IL-10,IL-12,tumor necrosis factor-α(TNF-α)and lncRNA XIST and miR-124 in peripheral blood mononuclear cells were detected in the three groups.The correlation between lncRNA XIST and miR-124 in acute stage was analyzed by Pearson method.The diagnostic value of lncRNA XIST and miR-124 in acute COPD was analyzed by receiver operating characteristic(ROC)curve.Multivariate logistic regression was used to analyze the influencing factors of acute COPD.Results Compared with the control group,IL-1β,IL-12,TNF-α and lncRNA XIST levels were significantly increased in the acute stage group and the stable stage group,and IL-10 and miR-124 expression levels were significantly decreased,the differences were statistically significant(F=58.987,95.674,106.138,237.468,45.710,126.014;all P<0.001).Correlation analysis by Pearson method showed that there was a significant negative correlation between lncRNA XIST and miR-124 in the acute stage group(r=-0.519,P<0.001).The area under curve(AUC)showed that the AUC of combined diagnosis was significantly larger than that of lncRNA XIST alone and miR-124 alone(Z=3.726,2.812;all P<0.05).Multiple logistic regression analysis showed that lncRNA XIST and miR-124 were influential factors in the acute exacerbation of COPD(all P<0.05).Conclusions The expression level of lncRNA XIST in peripheral blood mononuclear cells of patients with acute COPD was significantly increased,and miR-124 was significantly decreased,both of which had a negative correlation and were also influential factors in the acute COPD episode.The combined detection of both of them provided great help for the clinical diagnosis of COPD acute episode.
