摘要
目的 探讨宫颈癌合并人乳头瘤病毒(HPV)感染患者血清脂质运载蛋白2(LCN2)、长链非编码RNA-OIS1(lncRNA-OIS1)、微小RNA-3607-3p(miR-3607-3p)的表达水平及意义.方法 选取2017年5月-2022年5月武汉市第三医院收治的80例宫颈癌患者为观察组,70例宫颈上皮内瘤变患者为对照组.比较两组血清LCN2、lncRNA-OIS1、miR-3607-3p表达水平,以及HPV病毒载量、HPV阳性表达率.采用Pearson相关分析法分析宫颈癌患者血清LCN2、lncRNA-OIS1、miR-3607-3p与HPV病毒载量、HPV阳性表达率的相关性,采用多因素logistic回归分析影响宫颈癌合并HPV感染发病的危险因素.结果 观察组血清LCN2水平、HPV病毒载量、HPV阳性表达率分别为(84.01±18.47)pg/mL、(1 171.40±232.33)RLU/CO、100.00%,均明显高于对照组的(43.25±12.83)pg/mL、(669.50±100.32)RLU/CO、45.71%,lncRNA-OIS1、miR-3607-3p 水平分别为(0.70±0.15)、(0.33±0.10),均明显低于对照组(0.83±0.17)、(0.65±0.14),差异均有统计学意义(t/x2=15.480、16.753、58.163、4.976、16.251,P均<0.05).经Pearson相关分析结果显示,血清LCN2水平与HPV病毒载量、HPV阳性表达率均成正相关(r=0.515、0.430,P均<0.05),lncRNA-OIS1、miR-3607-3p 水平与 HPV 病毒载量、HPV 阳性表达率均成负相关(r=-0.600、-0.502、-0.587、-0.459,P均<0.05).Logistic回归性分析结果显示,血清LCN2是影响宫颈癌合并HPV感染发病的危险因素,血清lncRNA-OIS1、miR-3607-3p是发生宫颈癌合并HPV感染的保护因素,差异均有统计学意义(P均<0.05).结论 宫颈癌合并HPV感染患者血清LCN2明显高表达,lncRNA-OIS 1、miR-3607-3p明显低表达,且与HPV病毒载量、HPV阳性表达率密切相关.
Abstract
Objective To investigate the expression and significance of serum lipid carrier protein 2(LCN2),long non-coding RNA-OIS1(lncRNA-OIS1)and micrornA-3607-3p(miR-3607-3p)in patients with cervical cancer complicated with human papillomavirus(HPV)infection.Methods A total of 80 patients with cervical cancer admitted to the Third Hospital of Wuhan from May 2017 to May 2022 were selected as observation group,and 70 patients with cervical intraepithelial neoplasia were selected as control group.The levels of serum LCN2,lncRNA-OIS1,miR-3607-3p and HPV viral load and HPV positive expression rate were compared between the two groups.Pearson correlation analysis was used to analyze the correlation between serum LCN2,lncRNA-OIS1,miR-3607-3p and HPV viral load,HPV positive expression rate in patients with cervical cancer;multiple logistic regression was used to analyze the risk factors affecting the incidence of cervical cancer combined with HPV infection.Results The serum LCN2 level,HPV viral load and HPV positive expression rate in the observation group were(84.01±18.47)pg/mL,(1 171.40±232.33)RLU/CO and 100.00%,respectively,which were significantly higher than those in the control group[(43.25±12.83)pg/mL,(669.50±100.32)RLU/CO,45.71%];the levels of lncRNA-OIS1 and miR-3607-3p were(0.70±0.15)and(0.33±0.10),respectively,which were significantly lower than those in the control group[(0.83±0.17,(0.65±0.14)];the differences were statistically significant(t/x2=15.480,16.753,58.163,4.976,16.251;all P<0.05).Pearson correlation analysis showed that serum LCN2 level was positively correlated with HPV viral load and HPV positive expression rate(r=0.515,0.430,both P<0.05),while lncRNA-OIS1 and miR-3607-3p levels were negatively correlated with HPV viral load and HPV positive expression rate(r=-0.600,-0.502;-0.581,-0.459;all P<0.05.Logistic regression analysis showed that serum LCN2 was a risk factor for cervical cancer complicated with HPV infection,and serum lncRNA-OIS1 and miR-3607-3p were protective factors for cervical cancer complicated with HPV infection;the differences were statistically significant(all P<0.05).Conclusion In patients with cervical cancer complicated with HPV infection,serum LCN2 had significantly high expression level,while lncRNA-OIS1 and miR-3607-3p had significantly low expression levels,which were closely related to HPV viral load and HPV positive expression rate.
