Expression and clinical significance of miR-142-3p and miR-497-5p in serum of pregnant women with diabetes
Objective To investigate the expression and clinical significance of miR-142-3p and miR-497-5p in the serum of women with gestational diabetes mellitus(GDM).Methods A total of 142 pregnant women with GDM who underwent prenatal examinations in the Fifth Clinical College Medical Henan University of Chinese Medicine obstetrics and gynecology department from June 2019 to June 2022 were collected as research subjects(GDM group);in addition,142 pregnant women with normal glucose tolerance who matched the GDM group with general data were randomly selected as the control group.The serum levels of miR-142-3p and miR-497-5p were measured and compared between the two groups.Receiver operating characteristic(ROC)curve was applied to analyze the value of serum miR-142-3p and miR-497-5p levels in evaluating GDM in pregnant women.Results Compared with the control group,the level of miR-142-3p,the proportion of patients with diabetes history,the proportion of multiparous woman,fasting blood glucose(FPG),2 h postprandial blood glucose(2 h PG),fasting insulin(FINS),glycosylated hemoglobin(HbA1c),and insulin resistance index(HOMA-IR)in GDM group were obviously higher,the level of serum miR-497-5p was obviously lower,the differences were statistically significant(t=8.303,6.818,9.551,8.781,9.054,6.464,9.475,11.671,7.883;all P<0.05).The area under the ROC curve(AUC)of the combined detection of miR-142-3p and miR-497-5p for predicting GDM in pregnant women was 0.838,with a sensitivity of 73.24%,and a specificity of 82.39%,which was superior to their respective independent diagnosis(Z=4.379,3.170;all P<0.01).Conclusions The expression of miR-142-3p in serum of patients with GDM was obviously up-regulated,and the expression of miR-497-5p was obviously down-regulated.The combination of the two had a good reference value in predicting the occurrence of GDM in pregnant women.
