蒙古冰草全长转录组分析及参与木质素合成和干旱响应的羟化酶基因鉴定

蒙古冰草(Agropyron mongolicum)是广泛分布于我国北方荒漠草原地区的优质牧草,对寒冷、干旱、盐碱等环境胁迫具有很强的耐受性,具有很高的生态、经济和育种价值.然而,蒙古冰草基因组和转录组数据信息的缺乏严重限制了蒙古冰草的分子育种和开发利用等工作.本研究利用PacBio三代测序平台对蒙古冰草进行测序研究,获得了高质量全长转录组信息.对测序结果的组装获得了267560 个全长转录本,从中筛选出91855 条Unigene并注释了其中的83125 个.更进一步地从蒙古冰草全长转录组中挖掘了可能参与木质素合成的羟基转移酶基因.对这些基因在不同组织中以及干旱胁迫下的表达模式的研究表明,这些基因可能在木质素合成和干旱胁迫响应中发挥重要功能.本研究提供了蒙古冰草的高质量全长转录组资源,为挖掘蒙古冰草的优异基因资源提供了便利条件.
Full length transcriptome analysis of Agropyron mongolicum and identification of hydroxylase genes involved in lignin synthesis and drought response
Agropyron mongolicum is a high-quality forage widely distributed in the desert grassland areas of northern China.It has strong tolerance to environmental stresses such as cold,drought,and salinity,and has high ecological,economic,and breeding value.However,the lack of genomic and transcriptome data information on Agropyron mongolicum severely limits its molecular breeding and utiliza-tion.This study utilized the PacBio third-generation sequencing platform to sequence Agropyron mongolicum and obtained high-quality full-length transcriptome information.The assembly of sequencing results obtained 267 560 full-length transcripts,from which 91 855 Unigenes were obtained and 83125 of them were annotated.Furthermore,we identified hydroxylase genes that may be involved in lignin synthesis from the obtained full-length transcriptome of Agropyron mongolicum.Research on the expression patterns of these genes in dif-ferent tissues and under drought stress suggests that these genes may play important roles in lignin synthesis and drought stress response.Our study obtained high-quality full-length transcriptome resources of Agropyron mongolicum,benefiting the screening of gene resources for molecular breeding from Agropyron mongolicum.
