Gene Sequences of CO Ⅰ and Cyt b of Ardea insignis and Molecular Phylogenetic Analysis of Genus Ardea
Ardea insignis is a rare species and listed as a critically endangered species in the IUCN red list of threatened species.It belongs to genus Ardea of family Ardeidae.There are very few studies carried on this species.We obtained CO Ⅰ and Cyt b gene sequences of A.insignis using PCR technology.The length of the Cyt b gene was 1 163 bp,and the A + T content (51.7%) was higher than that of C + G (48.3%).The length of the CO Ⅰ gene was 722 bp,and the A + T content (52.5%) was also higher than that of C + G (47.5%).The third site of both genes showed the most remarkable base bias.Combined with the sequences of the other 6 Ardea species from GenBank,the phylogenetic tree of genus Ardea was constructed.The result showed that 7 species of genus Ardea formed a monophyletic clade with the posterior support at 96%.A.alba and A.insignis were found at the basal position of this clade,indicating their early diverge.Based on the genetic analysis,A.purpurea was considered to be the most closely related species to A.insignis.
