Genetic Diversity Analysis of Cultrichthys erythropterus in Four River Systems in Henan Province Based on cyt b Gene
In order to explore the genetic diversity and analyze the genetic structure of Cultrichthys erythropterus popula-tion in complex river systems in Henan Province,100 samples of C.erythropterus were collected from the four major river systems of the Yangtze River,the Yellow River,the Huaihe River and the Haihe River.The genetic diversity,genetic structure and historical dynamics of the population were analyzed based on mitochondrial cyt b gene.The results showed that the haplotype polymorphism(Hd)and nucleotide diversity(π)were 0.909 and 0.004 88,respectively,indicating high genetic diversity of the population.Unique haplotypes were identified in the four river systems,this indicated that the genetic differentiation between C.erythropterus populations is relatively big and the degree of genetic differentiation is rela-tively high.Phylogenetic trees,Bayesian phylogenetic trees and haplotype network maps were constructed and analyzed.It was found that there was no obvious phylogenetic differentiation and geographical structure among C.erythropterus popula-tions in the four river systems of Henan Province.AMOVA analysis showed that the variation within populations accounted for 72.11%,and the variation among populations accounted for 4.63%,indicating that the variation mainly occurred within populations.The results of Tajima's D and Fu's FS analysis showed that there was no recent population expansion event of C.erythropterus in Henan Province.This study provides reference for the protection and rational development of the germplasm resources of C.erythropterus.
