Culture in vitro and Plant Regeneration from Roots of Citrus paradisi Macf.Seedlings
Objective]The objective of the study was to establish regeneration system of C.para-disi in vitro and to provide useful references for the application of genetic transformation tech-nique.[Method]The roots of aseptic seedlings germinated from the seeds of 10 grapefruit varie-ties were cultured in vitro to obtain regeneration plants in the medium with different hormones and concentrations.[Results]The callus were observed in the medium MS+0.5 mg/L 6-BA+0.5 mg/L NAA and Perlist had the highest callus induction frequency (87.95%).The differentia-tion of adventitious buds was observed in the medium MS+2.0 mg/L 6-BA+0.5 mg/L NAA and Star Ruby had the largest differentiation rate of adventitious buds (37.84%).The regeneration buds were multiplied in the medium MS+2.0 mg/L 6-BA+0.2 mg/L NAA.The adventitious roots were induced in the medium 1/2 MS +1.0 mg/L IBA and the regeneration plants finally were obtained.The survival rates of regenerative plant ranged from 75.47% to 88.40%.[Con-clusion]The regeneration systems of 6 grapefruit varieties were established based on the study.
