Study on the Cold Demand of Buds of Three Plum Cultivars(Lines)and Expression Analysis of Dormancy-Related DAM Genes
[Objective]This study is to investigate the difference between bud dormancy and cold demand in three Prunus cultivars and the relationship between DAM gene expression and bud dormancy.[Method]The cold demand and germination rate of three plum cultivars(lines)were investigated,the DAM gene of plum was identified using bioinformatics method,and the expression of the DAM gene of the three varieties(lines)of plum was analyzed at different dormant stages of bud using qRT-PCR.[Result]The cooling requirements of"Wanshuqiangcuili"under the 7.2℃model,0~7.2℃model and Utah model were higher than those of'Qiangcuili'and'Cuihongli',and the cooling requirements of'Qiangcuili'and'Cuihongli'under the three statistical models of cold demand were the same.Six DAM genes(PdDAM1,PdDAM2,PdDAM3,PdDAM4,PdDAM5,PdDAM6)were identified,but the high-expression genes in the dormancy period and the internal dormancy release period(cold interval)in flower buds and leaf buds was DAM5,and the relative expression of DAM genes decreased during the dormant release period.[Conclusion]The difference of cooling requirements between flower buds and leaf buds of the three plum cultivars(lines)was revealed,and the DAM gene most related to sleep release was screened.These results provided a theoretical basis for the regulation of plum bud dormancy and ripening period.
