低温胁迫下华山松不同无性系幼苗抗寒性评价

[目的]为加速华山松抗寒育种进程,选育抗寒品种,提高栽培效率。[方法]以楚雄市紫溪山国家华山松良种基地10个优良无性系苗期叶片为材料,采用人工控温法对各无性系华山松幼苗进行低温处理,低温温度设置为4、0和-4℃,对照(CK)温度为常温25℃。低温胁迫72 h后测定其叶片的叶绿素含量、抗氧化酶活性、渗透调节物质以及丙二醛含量等生理生化参数的动态变化,运用主成分法和隶属函数法对无性系抗寒性进行综合评价。[结果]华山松各无性系幼苗在不同温度处理下各指标的变幅因种而异。随着温度的降低,华山松不同无性系幼苗叶片生理生化参数的动态变化差异显著,叶绿素含量整体呈下降趋势;超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性随着温度的降低均呈先上升后降低的变化趋势,但不同无性系变化幅度不同。可溶性蛋白质、可溶性糖、游离脯氨酸和丙二醛(MDA)含量逐渐增加。通过主成分分析,华山松不同无性系幼苗的耐寒性主要是由保护酶系统和渗透调节物质共同作用。[结论]利用隶属函数法对10个无性系综合分析得到其耐寒性强弱顺序依次为:7号>3号>1号>2号>8号>10号>5号>9号>6号>4号。
Evaluation on Cold Resistance of Different Clonal Seedlings of Pinus armandi Franch.under Low Temperature Stress
[Objective]The aim of this study is to select cold-resistant varieties of Pinus armandi Franch.and enhance cultivation efficiency to advance its breeding process.[Method]Using leaves from 10 supe-rior clones of P.armandi at the seedling stage,subjected to controlled artificial low-temperature treat-ment,at the Zixi Mountain National Base of P.armandi in Chuxiong City,Yunnan Province.The low temperatures were set at 4℃,0℃,-4℃,with a reference temperature of 25℃under normal condi-tions.The dynamic variations in physiological and biochemical parameters,including chlorophyll con-tent,antioxidant enzyme activity,osmotic regulation substance,and malondialdehyde content,were measured in these leaves after 72 hours of low temperature stress.We employed principal component analysis and membership function analysis methods to assess the cold resistance of these clones.[Re-sult]The variation in each index of the clones seedlings of P.armandi under different temperature treat-ments varied among different species.Decreased temperature significantly influenced the dynamic changes of physiological and biochemical parameters in the leaves of different clonal seedlings of P.ar-mandi.As temperature decreases,a declining trend was observed in the chlorophyll content.The activi-ties of superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT)exhibited an initial increase followed by a decrease as temperature decreased.However,the magnitude of changes in these parameters varied among different clones.Conversely,the contents of soluble protein,soluble sugar,free proline,and malondialdehyde(MDA)gradually increased with decreasing temoerature.The principal component analysis showed that the cold tolerance of different clonal seedlings of P.armandi was primarily influ-enced by the protective enzyme system and osmoregulatory substances.[Conclusion]The comprehen-sive analysis of 10 clones using the membership function method indicates the following ranking of their cold resistance strength:NO.7>NO.3>NO.1>NO.2>NO.8>NO.10>NO.5>NO.9>NO.6>NO.4.
