Expression and Distribution of β3-AR in Perirenal Adipose Tissue of Yak Calves
[Objective]The aim of this study was to explore the regulatory role of β3-adrenergic receptor(β3-AR,ADRB3)in cold adaptation of calf yaks.[Method]Healthy male calf yaks of different ages(1,7 and 30 days)were selected.The expression and distribution of ADRB3 mRNA,β3-AR protein and the hormone-sensitive triglyceride lipase(HSL)mRNA in the perirenal brown and white adipose tissue of calf yaks were detected by immunohistochemical staining(IHC),Real-time fluorescence quantitative PCR(qRT-PCR),and Western blot(WB).[Result]The IHC results showed that the β3-AR was dis-tributed both in perirenal brown and white adipocytes of yak calves,and the positive expression intensity of β3-AR was significantly increased from day 1 to 30(P<0.05).The results of qRT-PCR showed that the mRNA expression level of ADRB3 and HSL were significantly increased from day 1 to 30(P<0.05).The re-sults of WB showed that the protein expression level of β3-AR were significantly increased from day 1 to 30(P<0.05).[Conclusion]ADRB3 mRNA and β3-AR protein were expressed in both perirenal brown and white adipocytes of yak calves,and its expression level in white adipose tissue were higher than that in brown adipose tissue,indicating that β3-AR plays an important regulatory role in nonshivering thermogen-esis(NST)and lipolysis in perirenal adipose tissue of yak calves.
