Construction of SSR Fingerprints and Genetic Diversity Analysis of 63 Potato Materials in Mountainous Regions of Sichuan Province
[Objective]This experiment aimed to study the genetic diversity of 63 potato backbone parent materials suitable for cultivation in the mountainous regions of Sichuan Province.[Method]We identified 208 primers with high polymorphism from published literature.DNA was extracted from the 63 potato samples and amplified using PCR.[Result]The analysis revealed that 168 primers produced amplified bands,145 primers exhibited polymorphism,and 7 primers were able to completely distinguish the tested materials.[Conclusion]Utilizing these 7 pairs of primers to construct SSR fingerprints,the genetic similarity coefficients among the 63 potato materials ranged from 0.48 to 0.98.At a genetic similarity coeffi-cient of 0.481,the tested materials were categorized into three groups:A,B and C.At a coefficient of 0.614,the materials were further divided into seven groups.This indicates that the tested materials are genetically dis-tant from one another,providing valuable insights for the genetic breeding of potato hybrid parents.
