Ameliorating of Scutellaria babarta Extracts on Histopathology and Glycogen Deposition and Related Transmitter in HPA Axis of T2DM Rats
[Objective]This study aimed to investigate the therapeutic effect of alcohol extracts from the Scutellaria babarta on type 2 diabetes mellitus(T2DM)and its associated hypothalamic-pituitary-adrenal axis(HPA axis)hyperfunction,to provide a safe and effective treatment for T2DM from the per-spective of HPA axis.[Method]A total of 120 4-week-old SPF Wistar male rats were selected and ran-domly divided into normal diet group(GD group,n=24),high-carbohydrate and high-fat diet group(H group,n=96).H group was injected with STZ to induced T2DM model,and was randomly divided into model control group(HD),low(SD-L),medium(SD-M)and high(SD-H)dose groups with 24.The GD and HD groups were administrated with sanitary water,and the SD group were administrated with Scutellaria alcohol extract,respectively.The body weight,FBG,INS and blood lipid metabolism indexes of rats in each group were assayed on the 5 d,10 d,20 d and 30 d of treatment,respectively.Finally,the histopathological structure and the amount of glycogen changes in HPA axis were observed by H.E and PAS staining at 30 days.The changes in number of α and β cells in pancreas was observed by alde-hyde fuchsine stain.The expression of 5-HT and GABA in PVN and VMH were observed by IHC.[Re-sult]Compared with GD group,the following changes occurred in HD group:① The body weight and INS levels of the rats were extremely significantly decreased,and FBG levels was extremely significantly increased.② The number of PVN,VMH-GE and VMH-GI nerve cells decreased and neuroglial cells in-creased in hypothalamus;The number of basophil cells in pituitary gland decreased;zona glomerulosa,zona fasciculata of adrenal gland and pancreatic islet atrophy.③The glycogen content in HPA axis was significantly or extremely significantly increased.④ Pancreatic β cells necrosis,α cells increased.⑤The expression of 5-HT and GABA in PVN and VMH-GI in hypothalamus was extremely significantly decreased.These pathological changes were turned for the better after Scutellaria babarta extracts treat-ment.[Conclusion]These results indicate that the ethanol extract of S.melabelia has a favorable thera-peutic effect on T2DM,which can relieve the abnormal glycogen deposition and the overactivation of gly-cogen in HPA axis complicated by T2DM.
