Effect of Polysaccharides from Terminalia Chebula and Rubia Cordifolia L.Processed Terminalia Chebula on Cyclophosphamide Induced Liver Damage in Yellow Feathered Quails Based on Transcriptomics
[Objective]Investigate the effects of Terminalia chebula Retz polysaccharide(TCP)and Ru-bia cordifolia L.processed Terminalia chebula Retz polysaccharide(RTCP)on the liver damage of yellow feathered quails induced by cyclophosphamide.[Method]Eighty 21-day-old yellow feathered quails were selected and randomly divided into 10 groups,with 8 in each group.The blank group,model group,TCP group and RTCP group(0.25,0.5,0.75,1.0 g/kg),the experiment period was7-day.The polysaccharide group was given the corresponding dose of polysaccharide according to the body weight per day.The blank and model groups were given an equal dose of physiological saline by gavage,except for gavage for 4-7 days.The model group and polysaccharide group were injected with cyclophosphamide into the chest muscle according to body weight,while the blank group was injected with an equal dose of physiological saline.At the end of the experiment,HE staining was performed to examine liver pathologi-cal changes,and transcriptome sequencing was performed to analyze liver gene expression.[Result]Compared with the pathological section results of model group,the polysaccharide group had reduced in-flammatory cell infiltration,and the degree of vacuolization.Moreover,the effect of the high-dose poly-saccharide group was better than that of the low-dose group.The transcriptome sequencing results show that there are 1 126 differentially expressed genes between the blank group and the model group,601 dif-ferentially expressed genes between the TCP and the model group,513 differentially expressed genes be-tween the RTCP and the model group.Results of differential expression gene enrichment analysis point out that polysaccharides were participated in the regulation of calcium,insulin,and TGF-β,ErbB,PPAR,apoptosis and other signaling pathways.[Conclusion]The TCP and RTCP have a certain allevi-ating effect on liver tissue damage caused by cyclophosphamide in yellow feathered quails,possibly by regulating calcium,insulin,and TGF-β,ErbB,PPAR,apoptosis and other signaling pathways play a role.
