Effects of Carbonate Alkalinity Stress on Growth,Antioxidant Capacity and Expression of Growth-related Genes in Usscuri Whitefish Coregonus ussuriensis
To explore effects of alkalinity stress on Usscuri whitefish Coregonus ussuriensis Berg,the growth performance,antioxidant capacities and expressions of insulin-like growth factor-1(IGF-1)/growth hormone(GH)system genes were investigated in the Usscuri whitefish with an initial average body weight of(60.43±2.45)g reared in a circulating temperature-controlled aquarium of 180 cm×60 cm×47 cm at water temperature of 18℃and exposed to sodium bicarbonate alkalinity of 0 mmol/L,5 mmol/L,10 mmol/L,20 mmol/L and 30 mmol/L in each alkalinity group,30 fish per alkalinity group,and three parallel groups(n=10)for 6 weeks.The results showed that the growth performance of C.ussuriensis was significantly decreased under the carbonate alkalinity 20 mmol/L and 30 mmol/L compared with that in the control group(P<0.05),with the survival rate of 100%.The maximal weight gain rate(87.85%)and specific growth ratio(1.50%),and the minimal feed conversion ratio(0.91)were observed at 10 mmol/L alkalinity(P<0.05).The activities of glutathione(GSH),superoxide dismutase(SOD),and catalase(CAT)and malondialdehyde(MDA)content in liver tissue were found significantly increased in the 20 mmol/L and 30 mmol/L alkalinity group than those in the control group(P<0.05),without significantly changes in the activities of the above three enzymes in the 5 mmol/L or 10 mmol/L alkalinity group(P>0.05).The expression level of the IGF-1/GH system genes was found to be significantly up-regulated at alkalinity of 5 and 10 mmol/L(P<0.05),andthe expression level of the IGF-1/GH system gene was significantly decreased at alkalinity of 20 mmol/L and 30 mmol/L(P<0.05),with the minimum at 30 mmol/L.It was found that Usscuri whitefish had specific adaptability to 30 mmol/L alkalinity,and delivered better growth performance under the carbonate alkalinity 5 mmol/L and 10 mmol/L.These findings indicate that Usscuri whitefish has certain tolerances to carbonate alkalinity and has a good growth performance in the 10 mmol/L alkalinity,although the fish is not conducive to the growth,with survival rate of 100%,at alkalinity of 20 mmol/L and 30 mmol/L.
