Observation of Collagen Fiber Staining in Organs and Tissues of Crisp Grass Carp(Ctenopharyngodon idella)
In order to study the characteristics of collagen fiber distribution in different organs of grass carp Ctenopharyngodon idella fed with broad beans In this experiment,Masson stained sections were made on the back muscles,abdominal muscles,heart,hepatopancreas,spleen,foregut,midgut and hindgut tissues of 7.0 kg crisped grass carp and 4.5 kg common grass carp,and the quantity and distribution of collagen fibers were analyzed.The results showed that collagen fibers in the organ tissue were dyed blue and muscle fibers were dyed red.The collagen fibers of the back muscle and abdominal muscle of crisped grass carp are stained more deeply and their distribution area is larger.The volume fraction of collagen fibers in the cross section of back muscle and abdominal muscle is 1.27 times and 2.09 times of that in grass carp,respectively,indicating that the muscle tissue of crisped grass carp has more collagen fibers.In the hepatopancreas,spleen and heart tissues of crisped grass carp,the collagen fibers are mainly distributed in the arterial and venous wall space in the portal area of hepatopancreas,the splenic cord and the heart connective tissue.A large number of collagen fibers in the smooth muscle of normal grass carp and the intestinal tissue of crisped grass carp are distributed among the smooth muscle cells.The volume fraction of collagen fibers in the front,middle and rear intestines of crisped grass carp is 2.16 times,1.68 times and 2.66 times of that in grass carp,respectively.Compared with muscle,hepatopancreas,spleen and heart,the gut contains significantly more collagen fibers.The longitudinal and transverse sections of organ tissues showed that the types of muscle cells in muscle(skeletal muscle cells),intestine(smooth muscle cells)and heart(cardiomyocytes)did not change,but the number and distribution of collagen fibers and the structure of connective tissue were changed.
