Sequence Characteristics and Correlation With Lipid Deposition of PPARγ Gene in Common Carp Cyprinus carpio
To provide a foundation for research on the regulatory mechanism of peroxisome proliferator activated receptor(PPARγ)on lipid deposition in common carp Cyprinus carpio.PPARγ gene was closed and tissue expression characteristics of PPARγ gene were investigated in common carp with body weight of(215.20±8.21)g by RACE and real-time fluorescence quantitative PCR.The results showed that the full-length cDNA of PPARγ gene was 3 077 bp,including a 233 bp 5'untranslated region,a 1 311 bp 3'untranslated region,and an open reading frame encoding 510 amino acids.The PPARγ protein consists of four functional domains,including A/B region,DNA binding domain(DBD),hinge domain,and the ligand-binding domain(LBD),with the maximal similarity between the PPARγ gene of common carp and that of crucian carp Carassius auratus(98.63%)and zebrafish Danio rerio(87.28%).It was found that PPARγ gene was expressed in all tissues,with the maximal relative level of expression in liver,followed by brain,and the minimal in spleen and heart.The PPARγ gene expression level was significantly higher in abdominal fat in high fat group than that in low fat group(P<0.05),and the PPARγ gene expression level of dorsal muscle was significantly higher in high fat group than that in low fat group(P<0.01).PPARγ gene was shown to be used as a candidate gene for lipid deposition in common carp.The finding provides reference for further analysis of the molecular mechanism of PPARγ gene regulating lipid deposition in common carp.
