Inhibitory Effect and Mechanism of Pomegranate Peel Aqueous Extract on Vibrio parahaemolyticus
The inhibitory activity and mechanism of pomegranate peel aqueous extract(PPAE)against Vibrio parahaemolyticus were investigated in order to provide theoretical basis for the prevention and treatment of vibriosis in aquaculture.The minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC)of PPAE against V.parahaemolyticus were measured by double dilution method,and the relative conductivity was detected by conductivity meter.The leakage of nucleic acid and the biofilm inhibition rate of V.parahaemolyticus were determined by spectrophotometry,the protein synthesis of V.parahaemolyticus was detected by protein electrophoresis,the submicrostructure of V.parahaemolyticus was observed by scanning electron microscope,and the integrity of cell membrane in V.parahaemolyticus was analyzed by propidium iodide(PI)staining.The results showed that PPAE had obvious inhibitory effect on V.parahaemolyticus,with the MIC of 1.563 mg·mL-1 and MBC of 3.125 mg·mL-1,indicating that PPAE significantly inhibits the biofilm formation of V.parahaemolyticus(P<0.05).After PPAE treatment,nucleic acid leakage and relative conductivity of V.parahaemolyticus wereshown to be increased significantly(P<0.01),and the protein synthesis of Vibrio parahaemolyticus was found to be significantly inhibited.PPAE obviously changed the submicroscopic morphology and destroyed the cell membrane integrity of V.parahaemolyticus.
