Genetic Diversity Analysis of Coilia nasus in Lake Nansi and Lake Dongping Inferred from Mitochondrial D-loop Region
To explore the genetic diversity and phylogenetic relationship of Coilia nasus in Lake Nasi(34° 26'-35° 20'N,116° 34'-117° 21'E)and Lake Dongping(35° 43'-36° 7'N,116° 2'-116° 20'E),the mitochondrial control region(D-loop)sequence of C.nasus was analyzed in the two lakes.The results showed that a total of 65 mutation sites and 31 haplotypes were detected in 77 individuals.The haplotype diversity of the populations was found to be 0.805 7 and the nucleotide diversity to be 0.005 9 in Lake Nansi;the haplotype diversity of the populations was 0.819 3,and the nucleotide diversity was 0.011 7 in Lake Dongping.The Fst and subvariance analysis(AMOVA)showed that the genetic differentiation among the populations was moderate,and the molecular variation mainly existed within the populations,without significance in the genetic differences among the populations.Phylogenetic analysis showed that the populations clustered into two large branches,9 individuals from Lake Dongping being clustered into one branch,and most other individuals being clustered into one branch,which did not show obvious correspondence between geographical location and individuals.It is speculated that a few individuals in Lake Dongping have distant relatives,which may have gene exchange with other ecological types.
