Effects of DEHP on Apoptosis and Expression of Growth Related Genes of Zebrafish in Early Developmental Stage
To investigate the effect of diethylhexyl phthalate(DEHP)on apoptosis and expression of growth related genes of zebrafish in early developmental stage,freshly fertilized zebrafish embryos were placed in Petri dishes(φ 15 cm,200 eggs/dish)and exposed to solutions with DEHP mass concentrations of 0 µg/L(control group),10 μg/L,30 μg/L,and 300 μg/L for 4 d,with 5 parallel per group,and incubated in an incubator at water temperature of 28.5 ℃,exchange the culture medium every day,sucking out dead embryos and larvae in time,recording the number of fry in each dish.Ten fish larvae were randomly sampled for detection of apoptosis in vivo.The apoptosis of zebrafish larvae and the expression of growth related genes were analyzed by AO staining,apoptotic DNA ladder and real-time fluorescence quantitative PCR.The results showed that there was decrease to some extent in hatching rate and cell apoptosis in all three treatment groups compared with that in the control group(P>0.05).The AO staining fluorescence intensity was shown to be significantly higher in 30 µg/L and 300 μg/L DEHP groups than that in the control group(P<0.05).The expression levels of IGF1a and Ghra genes were found to be significantly decreased in zebrafish larvae in the three DEHP treatment groups(P<0.05).The expression level of IGF2b gene was significantly decreased in 30 µg/L and 300 μg/L DEHP treatment groups(P<0.05),and the expression of GIF1b gene was significantly down regulated in the 300 μg/L DEHP treatment group(P<0.05).The findings indicated that high concentration of DEHP induced apoptosis and inhibited the expression of growth related genes in zebrafish at the early development stage.
