DEHP对斑马鱼早期发育细胞凋亡及生长相关基因表达的影响

为探究邻苯二甲酸二乙基已酯(DEHP)对斑马鱼(Danio rerio)早期发育阶段细胞凋亡及生长相关基因表达的影响,将刚受精的斑马鱼胚胎放入培养皿(φ15 cm,200粒卵/皿)中,暴露于质量浓度分别为0 µg/L(对照组)、10 μg/L、30 µg/L和300 μg/L的DEHP溶液中4 d,每组5个平行.在28.5℃培养箱中孵化,每天更换培养液,及时吸除死亡胚胎和仔鱼,记录各皿中的鱼苗数,随机取10尾鱼活体检测细胞凋亡.利用AO染色、细胞凋亡DNA Ladder以及实时荧光定量PCR技术分析斑马鱼仔鱼的细胞凋亡及其生长相关基因的表达.结果显示,3个处理组的孵化率均低于对照组(P>0.05),出现不同程度的细胞凋亡,30 μg/L和300 µg/L组的AO染色荧光强度显著高于对照组(P<0.05).与对照组相比,3个处理组斑马鱼仔鱼IGF1a和Ghra基因的表达量均显著下调(P<0.05);30 μg/L和300 μg/L组IGF2b基因的表达量显著下调(P<0.05);30 μg/L组IGF1b基因的表达量显著下调(P<0.05).结果表明,较高浓度的DEHP能够诱导斑马鱼早期发育阶段细胞凋亡,抑制其生长相关基因的表达.
Effects of DEHP on Apoptosis and Expression of Growth Related Genes of Zebrafish in Early Developmental Stage
To investigate the effect of diethylhexyl phthalate(DEHP)on apoptosis and expression of growth related genes of zebrafish in early developmental stage,freshly fertilized zebrafish embryos were placed in Petri dishes(φ 15 cm,200 eggs/dish)and exposed to solutions with DEHP mass concentrations of 0 µg/L(control group),10 μg/L,30 μg/L,and 300 μg/L for 4 d,with 5 parallel per group,and incubated in an incubator at water temperature of 28.5 ℃,exchange the culture medium every day,sucking out dead embryos and larvae in time,recording the number of fry in each dish.Ten fish larvae were randomly sampled for detection of apoptosis in vivo.The apoptosis of zebrafish larvae and the expression of growth related genes were analyzed by AO staining,apoptotic DNA ladder and real-time fluorescence quantitative PCR.The results showed that there was decrease to some extent in hatching rate and cell apoptosis in all three treatment groups compared with that in the control group(P>0.05).The AO staining fluorescence intensity was shown to be significantly higher in 30 µg/L and 300 μg/L DEHP groups than that in the control group(P<0.05).The expression levels of IGF1a and Ghra genes were found to be significantly decreased in zebrafish larvae in the three DEHP treatment groups(P<0.05).The expression level of IGF2b gene was significantly decreased in 30 µg/L and 300 μg/L DEHP treatment groups(P<0.05),and the expression of GIF1b gene was significantly down regulated in the 300 μg/L DEHP treatment group(P<0.05).The findings indicated that high concentration of DEHP induced apoptosis and inhibited the expression of growth related genes in zebrafish at the early development stage.
